Agarases are hydrolytic enzymes that act on the hydrolysis of agar and have a broad range of applications in food, cosmetics and pharmaceutical industries. In this study, a glycerol feeding strategy based on induction mode optimization for high cell density and β-agarase production was established, which could effectively control acetate yield. First, exponential feeding strategy of glycerol with different overall specific growth rates(μ) was applied in the pre-induction phase. The results showed that the low μ(μ=0.2) was suggested to be the optimal for cell growth and β-agarase production. Second, the effects of induction temperature and the inducer concentration on cell growth and β-agarase production were investigated in the post-induction phase. When induced by isopropyl-β-d-thiogalactoside(IPTG), the strategy of 0.8 mmol/L IPTG induction at 20℃ was found to be optimal for β-agarase production. When cultivation was induced by continuous lactose feeding strategy of 1.0 g/(L·h), the β-agarase activity reached 112.5 U/mL, which represented the highest β-agarase production to date.Furthermore, the β-agarase was capable of degrading G. lemaneiformis powder directly to produce neoagarooligosaccharide, and the hydrolysates were neoagarotetraose(NA4) and neoagarohexaose(NA6). The overall research may be useful for the industrial production and application of β-agarase. 相似文献
Starfish oocytes with intact germinal vesicles (GVs) were cut along desired planes with glass needles or ligated using silk
thread loops into two parts and allowed to mature in vitro, and inseminated. The experimental results showed that (1) only
the parts with GVs or partial GV contents (PGVCs) cleaved, those without any GV materials did not; but nucleated and non-nucleated
fragments cut from mature eggs were able to divide; (2) the development of animal parts of oocytes containing GVs or PGVCs
was like that of animal fragments of matured oocytes with female pronuclei; most of them gave rise to permanent blastulae,
and just a few formed ectodermal vesicles with a little primary mesenchyme; (3) a large part of vegetal fragments with GVs
or PGVCs, and the vegetal parts of mature eggs without female pronuclei developed into small but normal embryos; (4) the fragments
containing GVs or PGVCs obtained from the oocytes along a plane parallel to the animal-vegetal (A-V) axis developed as normally
as the halves (with or without female pronuclei) severed from mature eggs along the same axis. Based on the data above, it
was concluded that (1) the non-chromatin materials in the oocyte GVs are indispensable for successful fertilization and cleavage
of starfish eggs; (2) some factor (s) located asymmetrically in the vegetal hemispheres of starfish oocytes is (are) responsible
for formation of the archenteron and primary mesenchyme. It is evident from the above findings that the oocyte cytoplasm of
the starfish had already regionalized before the GV break-down.

The auriferous veins at Jinniushan occurs within the Jinniushan faulted zone in the Kunyushan Granite. Optical observation reveals that gold ore body formed during the main stage of hydrothermal activity. Detailed geothermometric studies of fluid inclusions from the veins show that the forming temperature ranges between 130℃ and 370℃ and the salinity is from 4.01 to 15.21 wvt percent NaCl. The ore-forming fluid is featured by low to moderate salinity, and low to moderate temperature. According to investigations of the values of vapor/liquid and temperatures of the ore-forming fluids, we propose that the boiling fluid inclusions exist in the main mineralization stages. Fluid boiling is suggested as a mechanism for the precipitation of gold from the hydrothermal fluid in the Jinniushan gold deposit. 相似文献
As well known, the methods of remote sensing and Bowen Ratio for retrieving surface flux are based on energy balance closure; however, in most cases, surface energy observed in experiment is lack of closure. There are two main causes for this: one is from the errors of the observation devices and the differences of their observational scale; the other lies in the effect of horizontal advection on the surface flux measurement. Therefore, it is very important to estimate the effects of horizontal advection quantitatively. Based on the local advection theory and the surface experiment, a model has been proposed for correcting the effect of horizontal advection on surface flux measurement, in which the relationship between the fetch of the measurement and pixel size for remote sensed data was considered. By means of numerical simulations, the sensitivities of the main parameters in the model and the scaling problems of horizontal advection were analyzed. At last, by using the observational data acquired in agricultural field with relatively homogeneous surface, the model was validated.
Tanlu Fault Zone (TFZ) is a large NE trending deep fracture system in East China and is about 2500 km long. It extends from the south of Lujiang to Tancheng, and passes through the Bohai Sea and continues to the north, where it branches into Yilan ?Shulan Fault and Dunhua ?Mishan Fault. TFZ is a long lasting huge deep - rooted lithospherical fault system with different characteristics in different parts and at different periods of its evolution. TFZ also controls the distribution and occur-rences of many superlarge, large, middle and small sized gold ore deposits, e. g. Jiapigou, Haigou, Xiaoxihancha and Ciwei-gou gold deposits in Northeast China; Linglong, Jiaojia, Sanshandao, Taishang, Xincheng, Rushan, and Pengjiakuang gold deposits in Jiaodong Peninsula of Shandong Province; and Guilaizhuang and Yinan in Western Shandong Province. 相似文献
